Yes, it's tough not to sound like a jerk saying this, but I do sometimes marvel at my own collection, highlighted by the fact that I have many of the cards mentioned in this thread.

My overall most wanted card is the Stan Musial RC: https://www.tcdb.com/ViewCard.cfm/sid/16/cid/3964/1948-Bowman-36-Stan-Musial But I'd be thrilled to acquire any of his cards made (or listed here) before that one, except the one that I already own (1947-66 Exhibits W461 - kneeling).
My most wanted card that is reasonable for me to actually buy or receive in a trade would be the 1979 O-Pee-Chee Ozzie Smith RC. It's now been over 13 years since I finally got the Topps RC that I'd wanted since I was a child, I still can't seem to chase down the OPC RC.

I have a collection of 56 different Tony Gwynn cards and I had to go look to see if I had that one for you, but unfortunately I don't. Must be a good one.

I have to echo the setiments shared earlier, I am blessed to have a number of cards mentioned on this thread. I also have to cheat, since my most wanted is numbered to 25. Jere are my top 5 wants, cards that have been on my list seemingly forever that I just can't find, maybe because they are inexpensive, certainly not because of rarity!

I have been blessed with a wife that enjoys the hobby with me, even though the cards do not hold any sentimental value for her. Because of that she doesn't say anything about any money I spend on the hobby and will spend it when I won't.
My list would be
Baseball 2018 Topps A & G Ronald Acuna I need 2 to complete that set and this is the main one
Football 1971 Topps Terry Bradshaw
Hockey 1979 Topps Wayne Gretzky. We went to a show in Canada a few years back and I saw a number of O-Pee-Chee versions but no Topps.
Nascar 1997 Dale Earnhardt Shark Bite Relic I keep looking on Ebay and will probably buy this one in the next few months

This is a cool topic - fun to see everyone's answers!
For me, it's the 2005 Topps All-Time Fan Favorites Kevin McReynolds Rainbow auto /10. I was pretty sure it didn't exist, because I hadn't seen it anywhere, ever, and then one popped up on eBay a few years ago as part of a big lot that went for way more than I was willing to pay. I've got almost every other McReynolds card (excluding 1/1s); but that's the one that has eluded me for years.
The 1968 Seaver/Ryan might be #2 on my list.
