Start with the portion of My Collection at http://www.tradingcarddb.com/FAQ.cfm
From there, enter your collection, your For Trade and your Want list. Though this can take some time to do, if you really want to actively trade on this site, it is time well spent. The advantage of doing this is that others who want to trade will quickly see if your lists are compatible. Since I did this, I have over 140 trades and the proposals keep coming, big and small.
One suggestion: even if you have 15 identical cards to trade, just add one in your For Trade list. If someone asks for it in trade, then add a second one while filling out the trade request. The Transaction system will transfer one out of your list automatically leaving the second one showing for trade. This avoids cluttering your Trade list to others looking to trade. The only exception to this would be for Vintage cards (pre-1970) that have various conditions (you can show the condition of the card - your opinion or professionally graded - when listing your cards by doing a detailed add or edit).
